As above, the gearbox is likely to suffer (it's a very common problem), the max torque rating is the same 320 as the engine produces (hence the name M32) , 6th gear is usually the one to go (a whine and movement of the gear lever are the main signs).
Many people do map their cars and never have an issue (though many with no extra force have theirs fail!).
Minimum you should do is replace the oil for something thicker over the OEM spec (Redline MT90 is a popular option), and overfill it slightly (lubrication for 6th is supplied by the oil splashing in the gearbox!!).
Definitely blank the EGR, as for the DPF, I would recommend it, though there is some discussion over the new MOT specs (which state it needs to be present, though you could just core it).
