No number plate = Definite fail.

Wheels sticking out beyond the wheel arches should also be a fail. To be legal, the top of the tyre shouldn't stick out further than the lip of the arch. Depends how strict the tester is.

Wheels sticking out past the bodywork isn't an MOT failure, although the police might have something to say on the subject... Cars can be MOT'd without a number plate also, on the chassis number for non registered imports, diplomatic vehicles etc.
